VAX-31 Phase 3 Win Lifts Vaxcyte; $1B Raise Dilutes

  • VAX-31 hits Phase 3 goals, stock jumps Vaxcyte's lead vaccine VAX-31 met all main goals in a late-stage trial, matching Pfizer's and Merck's shots and beating them on some strains. The stock soared 57% to a 52-week high. This success is the biggest driver, making approval and launch much more likely.

This is the key event that directly caused the stock's massive jump and changes the company's outlook.

  • $1B capital raise dilutes shareholders Vaxcyte launched $1 billion in stock and convertible notes to fund VAX-31 development and launch. The stock fell 3.7% as investors worried about dilution. While the cash strengthens the balance sheet, it spreads future profits over more shares, pressuring the price short-term.

This is a new event that directly impacts the stock price and capital structure.

  • Data timing moved up to late October Vaxcyte narrowed its VAX-31 Phase 3 OPUS-1 readout to late October, earlier than the prior fourth-quarter guidance. This accelerated catalyst timeline raised expectations and is a positive driver, as it brings clarity and potential near-term value inflection.

This is a new update that changes the timing of a major catalyst, influencing investor sentiment.

  • Key executive hires bolster launch prep Vaxcyte added Pfizer's former vaccines chief medical officer and Teva's former legal head. These hires bring deep experience in vaccine development and commercialization, strengthening the company's ability to execute on VAX-31's regulatory and launch plans, which supports the stock.

This is a new development that enhances the company's leadership and execution capabilities.

Vaxcyte Raises US$1b After Positive Phase 3 VAX-31 Data

Vaxcyte has raised roughly US$1b through concurrent common stock, pre funded warrant and convertible note offerings, following positive Phase 3 data for its VAX 31 pneumococcal vaccine candidate. The financing came after the Phase 3 OPUS 1 readout, and the company's shares have traded at US$66.96, with a 7 day share price return of 17.68% and a year to date share price gain of 44.12%. The 1 year total shareholder return of 64.36% and 5 year total shareholder return of 175.56% reflect momentum reinforced by the OPUS 1 update and the US$1b mix of equity and convertible note financing completed this week. Vaxcyte is currently trading at 3.6x book value, compared with 2x for the wider US Biotechs group and 9.3x for closer peers, while a widely followed narrative framework anchors fair value closer to $75 using a 7.51% discount rate. Completion of a dedicated large scale manufacturing facility with Lonza, build out of a high volume fill finish line in North Carolina and plans to build commercial supply ahead of a possible VAX-31 launch could support stronger net margins, though OPUS-2 or OPUS-3 data or higher than modeled manufacturing costs could reset expectations.

Vaxcyte Surges 32% on Positive Phase 3 Data for VAX-31 Pneumonia Vaccine

Vaxcyte shares jumped about 32% in Monday morning trading after the company reported positive phase 3 topline data for VAX-31, its 31-valent pneumococcal conjugate vaccine candidate. The OPUS-1 trial tested non-inferiority and immunogenicity of VAX-31 against Merck's Capvaxive and Pfizer's Prevnar 20 in patients aged 50 and over. For the 11 serotypes shared by the three shots, VAX-31 met all primary noninferiority assessments, and three serotypes unique to VAX-31 plus cross-reactive serotype 20B met the OPA GMR superiority criterion. Vaxcyte plans to release results from two other phase 3 trials, OPUS-2 and OPUS-3, in the first half of 2027, with a BLA eyed for the first half of 2028. Approved vaccines cover significantly fewer pneumonia strains than VAX-31: Capvaxive covers 21 strains and Prevnar 20 covers 20.

Kodiak, Celldex, Rapport and Vaxcyte Face Key Fall Clinical Readouts

Four clinical-stage biotech companies are approaching potentially important mid- to late-stage data readouts this fall, with outcomes that could reshape their pipelines and valuations. Kodiak Sciences is on track to report top-line data this month from the phase III DAYBREAK study evaluating Zenkuda (tarcocimab tedromer) and KSI-501 for treatment-naïve wet age-related macular degeneration against Regeneron's Eylea, targeting a roughly $15-billion anti-VEGF market. Celldex Therapeutics expects a September/October 2026 top-line readout from its phase III EMBARQ-CSU1 and EMBARQ-CSU2 studies of barzolvolimab in chronic spontaneous urticaria, with 1,939 patients enrolled and a planned regulatory submission in 2027. Rapport Therapeutics is targeting October 2026 for phase II results on RAP-219 in bipolar mania, while Vaxcyte expects top-line safety, tolerability and immunogenicity data from the phase III OPUS-1 study of its 31-valent pneumococcal conjugate vaccine VAX-31 by the end of October 2026. Among the four, Kodiak carries a Zacks Rank #4 (Sell), while Celldex, Rapport and Vaxcyte each hold a Zacks Rank #3 (Hold).

Vaxcyte Hires Pfizer and Teva Veterans, Moves VAX-31 Phase 3 Data to October

Vaxcyte announced two C-suite hires on September 3 and narrowed the timeline for its most important catalyst, saying it now expects topline safety, tolerability and immunogenicity data from the VAX-31 adult Phase 3 OPUS-1 trial by the end of October rather than sometime in the fourth quarter. Luis Jodar joins as Chief Medical Officer after 17 years at Pfizer, where he was Chief Medical Officer for Vaccines and Infectious Diseases and helped lead clinical development and medical affairs for Prevnar 13 and Prevnar 20, the pneumococcal vaccines VAX-31 is trying to displace. David McAvoy joins as Chief Legal Officer from Teva Pharmaceutical, where he ran a 235-person legal, compliance and government affairs organization as Executive Vice President and Chief Legal Officer, and previously served as General Counsel of Endocyte through its $2.1 billion acquisition by Novartis and spent 27 years at Eli Lilly. The OPUS-1 trial alone dosed 4,049 participants in a head-to-head comparison against Prevnar 20 and Capvaxive, part of a broader Phase 3 program that has dosed 6,191 adults in total, roughly 3,500 of them with VAX-31, and Vaxcyte held about $2.5 billion in cash, cash equivalents and investments as of June 30, 2026. The trial measures safety, tolerability and immunogenicity against Prevnar 20 and Capvaxive rather than a fresh efficacy claim, so the near-term thesis rests on VAX-31 clearing a noninferiority bar, and the company still has no approved product or product revenue, with OPUS-2 and OPUS-3 data and the VAX-31 infant Phase 2 study not expected until the first half of 2027 and the VAX-A1 Group A Strep candidate not guided to topline data until the second half of 2027.

Vaxcyte CFO Sells Shares to Cover Tax Withholding

Vaxcyte's President and CFO Andrew Guggenhime reported the disposition of 2,705 shares, valued at approximately $165,492, in a recent SEC Form 4 filing, but the transaction was not a discretionary sale—it was an automatic surrender of shares to satisfy tax withholding obligations upon the vesting of restricted stock units. Following the transaction, Guggenhime retains about 164,000 shares, including 102,471 held directly and 61,850 held indirectly via ALG 2025 Grat Holdings LLC, with a total value of $10.12 million based on the September 2 market close. Vaxcyte, a clinical-stage biotech with no revenue, held $2.51 billion in cash and investments at the end of June, up from $2.44 billion at the end of last year, and reported a six-month net loss of $604.9 million, with operating expenses reaching $302.8 million in the second quarter. The company is spending heavily on manufacturing for a commercial launch that depends on trial data for its lead candidate VAX-24, with results expected in the fourth quarter, and shares have returned 95% over the past year.

Vaxcyte narrows VAX-31 data timing to late October

Vaxcyte expects to report topline results from its pivotal Phase 3 OPUS-1 trial of the pneumococcal vaccine candidate VAX-31 by the end of October, narrowing its previous guidance for a fourth-quarter readout. The OPUS-1 study, which is part of a three-trial adult program involving 6,191 participants, will provide safety, tolerability, and immunogenicity data from a noninferiority study in adults. The company also appointed former Pfizer vaccine executive Luis Jodar as chief medical officer and David McAvoy, formerly of Teva Pharmaceutical Industries, as chief legal officer, as it prepares for a potential Biologics License Application and commercial launch. VAX-31 is the broadest-spectrum pneumococcal conjugate vaccine in clinical development, designed to protect against 31 bacterial serotypes, and results from the other two trials, OPUS-2 and OPUS-3, are expected in the first half of 2027.

Vaxcyte's Next Year Hinges on VAX-31 Trial Results

Vaxcyte reported second-quarter results that underscore its reliance on upcoming data for its lead pneumococcal vaccine candidate, VAX-31, with three Phase 3 trials now fully enrolled and the first major readout expected in the fourth quarter of 2026. The company's net loss for the quarter ended June 30 widened to $284.3 million from $166.6 million a year earlier, driven by higher R&D spending of $267.9 million and G&A costs of $34.9 million. Cash and investments stood at $2.5 billion as of June 30, up from $2.44 billion at the end of 2025. The board added Dr. Moncef Slaoui, former chief scientific advisor to Operation Warp Speed, and Dr. John Markels, former president of Merck's global vaccines business. Hedge fund ownership fell from 57 to 47 funds, while short interest remains at 10% of the float, reflecting a divided market outlook.

Vaxcyte Doses First Patient in Phase 1 Trial of Group A Strep Vaccine

Vaxcyte has dosed the first participant in its Phase 1 first-in-human clinical trial of VAX-A1, an experimental vaccine against Group A Streptococcus, a disease for which no approved vaccine currently exists. The trial will enroll 80 healthy adults aged 18 to 40 in Australia, a country with high rates of Group A Strep disease and established research networks. The study is structured in two stages, with an initial safety checkpoint after 12 participants before expanding to the full cohort. All participants will receive two doses of VAX-A1 or a placebo spaced about two months apart and will be monitored for six months after the final dose, testing low, mid, and high dose levels to balance immune response and tolerability. Vaxcyte estimates that Group A Strep causes 800 million illnesses globally each year, ranging from strep throat to life-threatening infections and rheumatic heart disease.
